Optimization of Whole Blood Genomic DNA Extraction Method in Patients with Abnormal White Blood Cell Count

Abstract:Objective To solve the problem that it is difficult to extract genomic DNA from blood samples in patients with abnormal white blood cell(WBC)count.Methods Blood samples required for human leukocyte antigen(HLA)genotyping were used as the research objects.Based on WBC count,the samples were divided into different groups:normal WBC count group[WBC count between(4-10)×109 L-1,Normal group],low WBC count group(WBC 4×109 L-1,LWBC group),and high WBC co groupunt group(WBC>10×109 L-1,HWBC group).The LWBC group was further divided into four groups:(1-4)×109 L-1(LWBC1 group),(0.5-1)×109 L-1(LWBC2 group),(0.1-0.5)×109 L-1(LWBC3 group),<0.1× 109 L-1(LWBC4 group).DNA extraction was carried out using column kits,and the procedure of sample treatment was optimized.The purity,concentration of the extracted DNA,and the accuracy of the subsequent genotyping test results were used as indicators to evaluate the optimization strategy.Results After method optimization,the purity of the extracted DNA in each group was maintained between 1.60 and 2.00,the mean concentration of each group was 54.33 μg·L-1(Normal group),51.50 μg·L-1(LWBC1 group),53.20 μg·L-1(LWBC2 group),49.80 μg·L-1(LWBC3 group),25.80 μg·L-1(LWBC4 group)and 54.0 μg·L-1(HWBC group),respectively.Two methods were used for subsequent HLA genotyping,both were successfully completed with consistent results.Conclusion The whole blood Genomic DNA extraction can be performed on patients with abnormal WBC counts through optimization.
