Construction of Key Gene Network and Prognostic Model of Esophageal Squamous Cell Carcinoma

Abstract:Objective To screen the key genes related to the pathogenesis and prognosis of esophageal squamous cell carcinoma(ESCC)by analyzing the data in the public database.Methods One microarray maps GSE26886 from NCBI-GEO database were collected,including 9 ESCC tissues and 19 normal tissues.Comprehensive bioinformatics methods were used to screen differential genes,and according to the gene function,including KEGG pathway and go enrichment analysis,the protein-protein interaction network was constructed.The prognosis of key genes in ESCC was evaluated by external data.Results There were 374 up-regulated and 606 down regulated genes in ESCC,which were mainly enriched in PI3K-AKT pathway and extracellular matrix.Survival analysis showed that MAL,TGM3,CRCT1,KRT78,SPINK7 and CLCA4 were related to the prognosis of ESCC.Conclusion The pathogenesis of ESCC may be caused by frequent cell proliferation through PI3K-AKT pathway and extracellular matrix degradation related to other pathways.Prognosis related models can predict the 1-year and 3-year overall survival of ESCC.
