Correlation Between miR-122a,NETs,HMGB1 and Inflammatory Response and Secondary Liver Injury in Children with Sepsis

Abstract:Objective To explore the association between microRNA-122a(miR-122a),neutrophil trapping nets(NETs),high mobility group protein 1(HMGB1),and inflammatory response and secondary liver injury in children with sepsis.Methods A total of 42 children with sepsis and secondary liver injury treated in the First Affiliated Hospital of Zhengzhou University from January 2022 to March 2024 were selected(secondary liver injury group),and 55 children with simple sepsis were selected(non secondary liver injury group),50 healthy children were selected as the healthy control group.The differences in miR-122a,NETs,HMGB1 and other indicators between the each groups were compared,and the differences in miR-122a,NETs,and HMGB1 between patients with different prognoses in the Secondary liver injury group were analyzed.Results The relative expression levels of miR-122a,NETs,HMGB1,alanine aminotransferase(ALT),total cholesterol(TBIL),white blood cell count(WBC),procalcitonin(PCT),C-reactive protein(CRP),and interleukin-6(IL-6)in secondary liver injury group were higher than non secondary liver injury group and health control group(P<0.05).The relative expression level of miR-122a in children with secondary liver injury was positively correlated with WBC,PCT,CRP,IL-6,ALT,and TBIL(P<0.05).NETs in children with secondary liver injury were positively correlated with WBC,PCT,CRP,IL-6,ALT,and TBIL(P<0.05).HMGB1 was positively correlated with WBC,PCT,CRP,IL-6,ALT,and TBIL in children with secondary liver injury(P<0.05).The relative expression levels of miR-122a,NETs,and HMGB1 in deceased children were significantly higher than those in surviving children(P<0.05).The areas under the receiver operating characteristic curve for predicting mortality in children with miR-122a,NETs,and HMGB1 levels were 0.693(95%CI:0.527-0.860),0.769(95%CI:0.628-0.910),and 0.833(95%CI:0.709-0.957),respectively,with P<0.05.Conclusion The elevated levels of miR-122a,NETs and HMGB1 in children with sepsis accompanied are correlated with inflammatory factors and liver function indicators,and have certain application value in predicting the prognosis of secondary liver injury children.
