Effects of Gestational Diabetes Mellitus on Gut Microbiota and Immune Function in Full-Term Neonates

Abstract:Objective To investigate the effects of gestational diabetes mellitus(GDM)on gut microbiota and immune function in full-term neonates.Methods A total of 35 full-term newborns of GDM mothers who were admitted to the Department of Neonatology in the First Affiliated Hospital of Xinxiang Medical University from January 2022 to December 2022 were selected as the experimental group,and 35 full-term newborns of normal mothers who were delivered in the First Affiliated Hospital of Xinxiang Medical University during the same period were selected as the control group.Fecal samples were collected on the 3rd and 14th after birth,and 16S rRNA high-throughput sequencing technology was used to detect the levels of Bifidobacterium and Lactobacillus in feces.Peripheral blood samples were collected on the 3rd and 14th after birth.Serum immunoglobulin(IgA,IgM,IgG,C3,C4),CD56+NK cells and T lymphocyte subsets(CD4+,CD3+,CD4+/CD8+)were measured.Results On the 3rd and 14th day of newborn birth,the level of Bifidobacteria and Lactobacillus in the experimental group was lower than that in the control group(P<0.05).In both groups,the level of Bifidobacteria and Lactobacillus on the 14th day of birth was higher than that on the 3rd day of birth(P<0.05).On the 3rd and 14th day of newborn birth,the IgG and C3 levels in the experimental group were lower than those in the control group(P<0.05),while the differences in IgA,IgM,and C4 levels were not statistically significant(P>0.05).The CD56+NK cells,CD4+,CD3+,CD4+/CD8+in the experimental group were lower than those in the control group(P<0.05).On the 14th day of birth in the experimental group,there was a positive correlation between Bifidobacterium and Lactobacillus with CD4+and CD4+/CD8+in neonatal venous blood,but no significant correlation with CD3+.Conclusion GDM can lead to the reduction of intestinal probiotics and the decrease of humoral and cellular immune function in full-term neonates,which may affect the normal growth and development of the body.
