Expression of Transmembrane Protein 9 in Cervical Cancer and Its Relationship with Prognosis

Abstract:Objective To explore the expression and prognostic significance of transmembrane protein 9(TMEM9)in cervical cancer.Methods The GSCA and GEPIA2 databases were used to analyze the relationship between TMEM9 expression and overall survival(OS),progression free survival(PFS),and disease free survival(DFS).Immunohistochemistry(IHC)was employed to detect the expression level of TMEM9 in cervical cancer tissues and adjacent tissues,and the relationship between TMEM9 expression and OS was analyzed.Western blotting(WB)was used to detect the protein expression level of TMEM9 in cervical cancer cells and normal cervical epithelial cells.Results The analysis results of the GSCA database showed that patients with high expression of TMEM9 had shorter OS(P=0.036)and PFS(P=0.003)compared to patients with low expression of TMEM9,with a median OS of about 90 months and a median PFS of about 80 months.The analysis results of GEPIA2 database showed that patients with high expression of TMEM9 had shorter OS(P=0.021)and DFS(P=0.005)compared to patients with low expression of TMEM9.The IHC results showed that the expression level of TMEM9 in cervical cancer tissues was higher than that in adjacent cervical tissues(P=0.042).TMEM9 was mainly expressed in the cytoplasm of cervical cancer tissues and adjacent tissues.The WB results showed that the expression level of TMEM9 protein in cervical cancer cells was higher than that in normal cervical epithelial cells.Conclusion Patients with high expression of TMEM9 have poor prognosis,and TMEM9 is expected to become a predictive factor for the prognosis of cervical cancer.
