Expression and Clinical Significance of NEDD8 and EGFR in Liver Tissues of Children with Biliary Atresia

Abstract:Objective To investigate the clinical significance of neural precursor cell expressed,developmentally down-regulated 8(NEDD8)and epidermal growth factor receptor(EGFR)expression in children with biliary atresia(BA).Methods Ninety-one children with BA admitted to First Affiliated Hospital of Xinxiang Medical College from February 2020 to July 2021 were selected as the research object(BA group),and 88 congenital choledochectasis children with normal liver function were selected as the control group during the same period.According to the Ohkuma's classification of liver fibrosis,children with BA were divided into class Ⅰ group with 21 cases,class Ⅱ group with 22 cases,class Ⅲ group with 28 cases,and class Ⅳ group with 20 cases.Quantitative real-time PCR(qRT-PCR)method was used to detect the levels of NEDD8 mRNA and EGFR mRNA in liver tissues,immunohistochemistry was used to detect the expression levels of NEDD8 and EGFR proteins in liver tissues,an automatic biochemical analyzer was used to detect the liver function indicators total bilirubin(TBIL),gamma glutamyl transpeptidase(GGT),aspartate aminotransferase(AST),alanine aminotransferase(ALT).Pearson method was used to analyze the correlation between NEDD8 mRNA,EGFR mRNA and liver function indexes in the liver tissues of BA children.Results Compared with the control group,the levels of NEDD8 mRNA,EGFR mRNA,TBIL,GGT,AST,ALT and the positive rates of NEDD8 and EGFR proteins in the BA group were higher,and the difference was statistically significant(P<0.05).NEDD8 mRNA and EGFR mRNA in liver tissues of BA children were positively correlated(r=0.615,P<0.05),and both were positively correlated with liver function indexes TBIL,GGT,AST and ALT(P<0.05).The positive rates of NEDD8,EGFR mRNAs and proteins in liver tissues of BA children increased with the aggravation of liver fibrosis.Conclusion The expression of NEDD8 and EGFR mRNAs and proteins may be related to children's BA and the degree of liver fibrosis.
