Correlation of Serum Levels of PINK1,Serotonin,AQP4 and Treatment Effect in Children with Epilepsy

Abstract:Objective To observe the changes of serum levels of PTEN induced putative kinase 1(PINK1),serotonin and aquaporin 4(AQP4)in children with epilepsy,and to analyze their correlation with the therapeutic effect of levetiracetam+valproate sodium and its predictive value for treatment failure.Methods A total of 90 cases of children with epilepsy from June 2020 to June 2023 were selected as the research objects,and they were divided into effective group(67 cases)and ineffective group(23 cases)according to the treatment effect after 3 months.The clinical data and the levels of serum PINK1,serotonin and AQP4 before and 1 month after treatment were compared between the two groups.Multivariate logistic regression analysis was used to analyze the influencing factors of treatment failure,the predictive value of serum levels of PINK1,serotonin and AQP4 after 1 month of treatment for treatment ineffectiveness after 3 months of treatment was evaluated.A decision curve was drawed to evaluate the level of serum PINK1,serotonin and AQP4 to predict the clinical net return rate of treatment failure.Results After treatment,the levels of serum PINK1 and serotonin in the ineffective group were lower than those in the effective group,and the level of serum AQP4 in the ineffective group was higher than that in the effective group(P<0.05).After one month of treatment,serum levels of PINK1 and serotonin were independent protective factors for treatment ineffectiveness,and serum AQP4 level was independent risk factors for treatment ineffectiveness(P<0.05).The area under the curve(AUC)of the combined prediction of serum levels of PINK1,serotonin and AQP4 after 1 month of treatment(after 3 months of treatment)was greater than that of the single index prediction(P<0.05).When the risk threshold was 0.1-0.8,the combined prediction of serum levels of PINK1,serotonin and AQP4 for the net return of ineffective treatment was better than that predicted of single index prediction.Conclusion The levels of serum PINK1,serotonin and AQP4 in children with epilepsy are closely related to the treatment effect,and the combined detection of their levels has a certain predictive value for the treatment failure.
