Bioinformatics Analysis of Programmed Cell Death Protein 1 in Pan-Cancer

Abstract:Objective To explore the bioinformatics characteristics of programmed cell death protein 1(PDCD1)in pan-cancer and its correlation with clinical prognosis.Methods The gene expression profile information and clinical information of 33 kinds of common tumors were downloaded from TCGA database,and the data were standardized.The correlation between PDCD1 gene expression and clinical survival information,immune cell infiltration,tumor mutation burden and microsatellite instability was analyzed.CIBERSOR immune score was used to study the correlation between infiltration of different types of immune cells and PDCD1 gene expression in pan-cancer.protein-protein interaction(PPI)network was constructed,and the protein coding genes closely related to PDCD1 coexpression were screened,and the KEGG and GO pathway enrichment analysis was carried out.The expression of cell death related genes(ferroptosis,cuproptosis,pyroptosis)were extracted from 33 kinds of tumors,and the correlation with PDCD1 gene co-expression was analyzed.Results The expression of PDCD1 gene in various types of tumor tissues was higher than that in normal tissues,and affected the overall survival,progression-free survival,disease-free survival and disease-specific survival of patients,and the differences were statistically significant.The expression of PDCD1 gene in pan-cancer was correlated with multiple immune cell infiltration,tumor mutation burden,microsatellite instability and so on.PDCD1 gene was co-expressed with cell death related genes(ferroptosis,cuproptosis,pyroptosis)in many types of tumors.Ten protein coding genes(PDCD1LG2,CD3D,CD247,HJA-DRB1,PTPN11,HLA-DQB1,CD4,CD80,CD274,CD3E)closely related to PDCD1 gene were screened.Enrichment analysis showed that these genes were closely related to the expression of PD-L1 expression and PD-1 checkpoint pathway in cancer and cell adhesion molecules pathways.Conclusion PDCD1 may be a gene target for many types of tumors.
