Impact of Seasonal Variations on Pre-and Post-thaw Donor Sperm Quality in Henan Province

Abstract：Objective To evaluate seasonal variation in the quality of pre-and post-thaw parameters and the progressive motility recovery rate among sperm bank donors from Henan human sperm bank,and to provide reference for the best season of semen cry opreservation.Methods A total of 7 946 semen samples donated at Henan human sperm bank between January 2019 and December 2021 were collected from 1 202 qualified donors.Routine semen analysis was performed,and semen meet the freezing standard were cryopreserved in aliquots and analyzed after thawing.Results The semen volume in autumn[(2.41±0.69)mL]was lower than in spring,summer and winter[(2.56±0.83),(2.53±0.78),(2.53±0.81)mL],the difference was statistically significant(P＜0.05).Sperm concentration was highest in spring[(62.87±2.31)× 106 mL-1],as compared with autumn,winter and summer[(62.65±2.06)× 106 mL-1,(62.48±1.95)× 106 mL-1,(62.34±1.73)× 106 mL-1],and the difference was statistically significant(P＜0.05).The percentage of sperms with progressive motility in spring[(62.45±1.15)％]and autumn[(62.46±1.04)％]was higher than winter[(62.29±1.04)％],the difference was statistically significant(P＜0.05),and summer[(62.35±1.01)％]was lower than autumn[(62.46±1.04)％],the difference was statistically significant(P＜0.05).Sperm concentration after frozen recovery in spring[(41.72±1.49)× 106 mL-1]and autumn[(41.83±1.39)× 106 mL-1]were higher than winter[(41.53±1.53)× 106 mL-1],the difference was statistically significant(P＜0.05),and summer[(41.7±1.48)× 106 mL-1]was lower than autumn[(41.83±1.39)× 106 mL-1],the difference was statistically significant(P＜0.05).The percentage of forward motile sperm after frozen recovery in autumn[(42.32±0.99)％]was higher than spring[(42.21±0.92)％],summer[(41.7±1.48)％]and winter[(42.21±1.04)％],the difference was statistically significant(P＜0.05).The progressive motility sperm recovery rate in winter[(67.77±1.4)％]and autumn[(67.76±1.25)％]was higher than summer[(67.61±1.32)％]and spring[(67.63±1.27)％],the difference was statistically significant(P＜0.05).Conclusion Seasonal factors have effects on the semen parameters and the progressive motility sperm recovery rate in Henan province.It is suggested that donating semen should be appropriate during seasons other than summer.
