Identification of Immune Infiltration-Related Biomarkers Involved in Hashimoto's Thyroiditis Based on GEO Database

Abstract:Objective This study attempts to find the key markers and potential molecular mechanisms of Hashimoto's thyroiditis(HT)from the perspective of bioinformatics,and provide new ideas for the therapeutic targets of HT.Methods Two datasets GSE138198 and GSE549586 were downloaded from the GEO database.A total of 13 HT samples and 10 healthy control samples were included in the study.The 1 618 immune-related genes were also collected.The differentially expressed gene analysis,gene ontology analysis,Kyoto gene and genome pathway enrichment analysis,protein interaction analysis,gene set enrichment analysis,LASSO regression analysis,consistent clustering analysis,immune infiltration analysis were gradually performed to identify the key genes.Results Total of 803 differential genes between case and control groups were screened,of which 538 were up-regulated and 265 were down-regulated.The 134 were immune-related genes.By protein interaction analysis and LASSO regression analysis,3 key genes including the TLR3,CCL5,and CXCR5 were identified.According to the expression levels of the three characteristic genes,the HT samples were divided into two subtypes by consistent cluster analysis.The gene expression level,immune infiltration level,and enriched pathways were significantly different between two subtypes.Conclusion The identified 3 key genes related to HT can favorably stratify the populations,which may be the therapeutic targets in the treatment of HT.
