Therapeutic Effect and Mechanism of Modified Chaihu Danggui Decoction on Liver Fibrosis Rat

Abstract:Objective To investigate the effect and mechanism of modified Chaihu Danggui Decoction in treating hepatic fibrosis(HF)rats.Methods Ten rats were randomly selected from 52 SD rats as the blank group,and the remaining rats were intraperitoneally injected with 40%CCl4 olive oil to construct HF models.After successful modeling,the remaining rats were randomly divided into the model group,the low-dose,medium-dose,and high-dose modified Chaihu Danggui Decoction groups.Rats'appearance was observed during the experiment,and the rats were sacrificed after treatment for sampling.Liver function and hyaluronidase(HA),laminin(LN),procollagen Ⅲ peptide(PⅢP)and type Ⅳ collagen(CⅣ)of HF in the rats were detected.The levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β),alpha-smooth muscle actin(α-SMA)and type Ⅰ collagen(COL-Ⅰ)in serum and liver tissues were measured by ELISA.The protein and mRNA expression of PI3K,AKT and Bel-2 in liver tissues was determined by Western Blot and qRT-PCR.Results After the end of experiment,serum levels of glutamic-pyruvic transaminase(GPT)and glutamic-oxaloacetic transaminase(GOT)in the model group were higher than those in the blank group,and serum albumin(ALB)level was lower than that in the blank group(P<0.05).Compared with the model group,serum levels of GPT and GOT were lower,and ALB level was higher in the three modified Chaihu Danggui Decoction groups(P<0.05).The difference between the high-dose group and the blank group was not statistically significant(P>0.05).Compared with the blank group,the levels of HA,LN,PⅢP and CⅣ were higher in the model group(P<0.05).Compared with the model group,the four indicators of HF were lower in the three modified Chaihu Danggui Decoction groups(P<0.05),but there was no statistical difference between the high-dose group and the blank group(P>0.05).The levels of TNF-α,IL-6 and IL-1 β in serum,the expression levels of SMA and COL-Ⅰ in liver tissues of the model group were higher than those in the blank group(P<0.05).The levels of TNF-α,IL-6 and IL-1 β in serum,the expression levels of SMA and COL-Ⅰ in liver tissues of the three modified Chaihu Danggui Decoction groups were lower than those in the model group(P<0.05),but there was no statistical difference between the high-dose group and the blank group(P>0.05).The protein and mRNA levels of PI3K,AKT and Bel-2 in liver tissues of the model group were higher than those in the blank group(P<0.05).The protein and mRNA levels of PI3K,AKT and Bel-2 in liver tissues of the three modified Chaihu Danggui Decoction groups were lower than those in the model group(P<0.05).There was no statistical difference between the high-dose group and the blank group(P>0.05).Conclusion Modified Chaihu Danggui Decoction may exert anti-HF effects through alleviating hepatic inflammatory injury,inhibiting the activity of PI3K/AKT/Bcl-2 signaling pathway,and reducing extracellular matrix deposition.
