Study on Biomarkers for Brain Injury Consciousness Disorder Based on Serum Metabolomics

Abstract:Objective Based on the means of metabolomics,the specific endogenous biomarkers of patients with consciousness disturbance caused by brain injury were identified to provide basis for the study of clinical diagnosis and treatment mechanism.Methods Blood samples were collected from patients with consciousness disorder treated for forebrain injury(disease group,35 cases)and healthy physical examination subjects(healthy control group,40 cases),ultra-high performance liquid chromatography-quadrupole-electrostatic field orbital trap mass spectrometry(UHPLC-Q-Orbitrap HRMS)technology was used.Combined with multivariate statistical analysis,the serum samples of the disease group and the healthy control group were analyzed by metabonomics comparison,metabolites that met both multivariate statistical analysis screening criteria(VIP value>1)and univariate statistical analysis screening criteria(P<0.05)were selected as biomarkers with significant differences,and the related metabolic pathways were found.Results Through the MetaboAnalyst online analysis platform,compared with the healthy control group,53 different metabolites such as creatine,carnitine and glutamate were obtained in the disease group,and 27 metabolic pathways with high correlation with potential biomarkers were screened out,and 10 pathways with P<0.05 were selected,which may be related to the occurrence,development and prognosis of brain injury consciousness disorder.Conclusion There are metabolic abnormalities in patients with brain injury consciousness disorder,and the metabolomics screening study of biomarkers provides a new strategy for the basic research of identifying endogenous metabolic disorders in patients with brain injury consciousness disorder,clinical diagnosis,treatment and prognosis.
