Correlation Between CCR7 and MCM10 Proteins in Primary Liver Cancer Tissue and Clinical Pathological Characteristics and Prognosis

Abstract:Objective To explore the correlation between chemokine receptor 7(CCR7)and microsomal maintenance protein 10(MCM10)in primary liver cancer(PHC)tissue and clinical pathological characteristics and prognosis.Methods A total of 90 patients with PHC in Zhoukou Central Hospital from January 2017 to December 2020 were selected as the study objects.All patients underwent hepatocellular carcinoma resection,and tumor tissues and adjacent normal tissues were taken during the operation.The expression of CCR7 and MCM10 protein in different PHC tissues was compared,x2 test was used to analyze the relationship between CCR7 and MCM10 expression and clinicopathological features,Kaplan-Meier analysis was used to analyze the relationship between CCR7 and MCM10 protein expression and prognosis of PHC patients,and Cox regression analysis was used to analyze the influencing factors of prognosis of PHC patients.Kaplan-Meier analysis was used to analyze the relationship between the expression subgroups of CCR7 and MCM10 protein and the prognosis of patients with PHC.Results The positive expressions of CCR7 and MCM10 protein in PHC patients were 66.67% and 75.56%,respectively,significantly higher than those in normal tissues(23.33% and 27.78%),with statistical significance(P<0.05).The positive expression rates of CCR7 and MCM10 protein in cancer tissues of patients with lymph node metastasis and vascular invasion(79.25%,94.34% and 81.58%,89.47%)were higher than those of patients without lymph node metastasis and vascular invasion(48.65%,48.65% and 55.77%,65.38%),the differences were statistically significant(P<0.05).Kaplan-Meier survival analysis showed that the 3-year survival rates of patients with positive expression of CCR7 protein and MCM10 protein were 39.66%(23/58)and 41.54%(27/65),respectively,which were significantly lower than those of patients with negative expression of CCR7 protein,68.97%(20/29)and 72.73%(16/22),the differences were statistically significant(P<0.05).Cox regression analysis showed that lymph node metastasis(95%CI:1.129-3.025),vascular invasion(95%CI:1.463-3.147),CCR7 protein(95%CI:1.407-3.524),MCM10 protein(95%CI:1.384-3.441)were independent risk factors for prognosis in patients with PHC(P<0.05).Kaplan-Meier analysis showed that CCR7 protein positive+MCM10 protein positive subgroup had the worst prognosis,and CCR7 protein negative+MCM10 protein negative subgroup had the best prognosis,with statistical significance among all subgroups(P<0.05).Conclusion The expression of CCR7 and MCM10 protein in PHC tissues is correlated with clinicopathological features and prognosis.Positive expression of both proteins is a risk factor for poor prognosis,which can be used as an auxiliary index for clinical evaluation and prognosis prediction,and has certain guiding significance for clinical prevention and treatment.
