The effects of Antinociceptive on rat model of osteoarthritis pain

Abstract:Objective To explore the effects of Antinociceptive on rat model of osteoarthritis pain.Methods All rats under anesthesia of 3% isoflurane and oxygen, iodophor was used for disinfection of the left knee.FCA-OA Group (n=10): patellar ligament injection of FCA (1 mg/ml, Mycobacterium tuberculosis, Sigma, UK) 150 μl into the left knee joint using micro-syringe (BD Microfine, USA).The control group (n=15): the same volume injection of saline solution into the left knee joint.Rats were given plenty of water and food.14 days after FCA injection, according to former study, there were stable OA pain behaviors in rats, so each rat was given 30 mg/kg LCM through the spinal skin, mechanical thresholds, thermal pain threshold, cold pain threshold were assessed before and 1 hour after LCM.Results Compared with the sham group, the mechanical threshold in OA group was lower in POD14, and the heat withdrawal latency were reduced in POD14, as well as cold withdrawal frequency was increased in POD14.Then compared with the no drug time point, 1 hour after the LCM, the mechanical threshold in OA group was increased, and the heat withdrawal latency was increased, as well as cold withdrawal frequency reduced, and all the differences were significant.Conclusion Animal studies have shown that injection of LCM can reduce mechanical threshold, heat, cold thresholds and pain in OA rats.
