Effect of DEC1 gene expression on proliferation and invasion abilities of human esophageal cancer ECA109 cells

Abstract:Objective To investigate the effect of DEC1 gene expression on proliferation and invasion abilities of human e-sophageal cancer ECA109 cells.Methods Human esophageal cancer ECA109 cells were selected and transfected with plasmid pc DNA3.1(-)/DEC1 (study group)and pcDNA3.1(-)(control group).The expression of DEC1 mRNA was evaluated by real -time PCR,and the protein of cyclin D1,MMP9 and DEC1 were evaluated by Western blot.The cell proliferation and inva-sion ability were evaluated by Transwell test,colony formation assay and CCK -8 assay.Results The expression levels of DEC1 mRNA and DEC1 in the study group were significantly higher than those in the control group(P <0.05),but the expression lev-els of cyclin D1 and MMP9 were significantly lower than that of the control group(P <0.05).The cell proliferation and invasion ability of the study group was lower than that of the control group(P <0.05).Conclusion The expression level of DEC1 may affect the proliferation and invasion of human esophageal cancer ECA109 cells,and the inhibition of cyclin D1 and MMP9 expres-sion may be the mechanism of its action.
