The efficacy of different doses of dexmedetomidine combined with sufentanil on postoperative analgesia in children undergoing hypospadias operation

Abstract:Objective To observe the efficacy of different doses of dexmedetomidine (Dex)combined with sufentanil on post-operative analgesia in children undergoing hypospadias operation.Methods 80 cases of children undergoing the elective hypospa-dias operation were randomly divided into 4 groups,20 cases in each group:group C received sufentanil 0.04 μg/(kg·h),group D1 received sufentanil 0.04 μg/(kg·h) +Dex 0.02 μg/(kg·h),group D2 received sufentanil 0.04 μg/(kg·h) + Dex 0.04 μg/(kg·h),group D3 received sufentanil 0.04 μg/(kg·h) +Dex 0.06 μg/(kg·h).4 groups used the same induc-tion and maintenance of anesthesia.Intravenous injection of sufentanil with 0.15 μg/kg was conducted at 30 min before the end of operation.The patient -controlled intravenous analgesia (PCIA)at the end of operation was immediately jointed.The non -invasive blood pressure,heart rate,SpO2 ,the score of faces pain scale (FPS)and Ramsay sedation of children at 2,4,8,12, 24 and 48 h after operation were recorded,and the press number of PCIA and the incidence of side effects were recorded.Re-sults Compared with group C,at each time point,FPS scores in group D1 ,D2 and D3 were lower (P <0.05),and they were lower in group D2 and D3 than in group D1 (P <0.05);Ramsay sedation scores were higher in group D1 ,D2 and D3 than in group C at all the time points (P <0.05),and they were positively correlated with Dex dose in 4 hours (P <0.05);Compared with group C and D1 ,the press number of PCIA and the incidence of nausea and vomiting in group D2 and D3 declined (P <0.05),but there were 2 cases of excessive sedation in group D3 .No bradycardia,hypotension and respiratory depression hap-pened in 4 groups.Conclusion In postoperative analgesia for children with hypospadias,Dex could enhance the effect of analge-sia and reduce side effects,but it needs to prevent excessive sedation.The dose of 0.04 μg/(kg·h)is the best selection.
