Efficacy of fresh -frozen plasma in the treatment of brain injury accompanied by hemorrhagic shock in rats

Abstract：Objective To investigate the efficacy of fresh -frozen plasma (FFP)on brain injury accompanied by hemorrhagic shock in rats.Methods 25 healthy SD rats were selected and FFP was prepared.108 healthy adult SD rats were randomly divided into three groups.The sham group (18 cases)only opened skull bone window,vehicle group and FFP group (45 cases in each) used Feeney improved method and Wiggers improved method to make rat brain injury and hemorrhagic shock model.After 60 mi-nutes,the vehicle group were given three times the amount of blood loss saline,FFP group were given the same amount of blood loss FFP.Blood -brain barrier permeability were measured by EB staining,cerebral edema were measured by dry -wet weight method.The improvement of neurological disorders were measured by mNSS score,and brain neuronal cell survival situation were deserved by Nissl staining on the 7th day.Results On the 1st,3rd,7th day,brain edema of FFP group was lower than vehicle group (P <0.05).In 12,24,72 h,EB content of FFP group was lower than the vehicle group (P <0.05),mNSS neurological scores of FFP group were lower than the vehicle group on the 3rd,7th day.Conclusion FFP could relieve brain edema,reduce the permeability of blood -brain barrier,improve nerve function in the treatment of brain injury and hemorrhagic shock resuscitation in rats.
