Analysis of clonal espression of T cells in metastatic lymph node of patients with breast cancer

Abstract:Objective To investigate the distribution and clonality of TCR BV subfamilies T cells in patients with breast cancer.Methods The CDR3 region genes of 24 TCR BV subfamilies were amplified by RT -PCR in T cells from 2 reactive hyperplasia lymph nodes and 10 patients from lymph node metastasis,and the CDR3 size lengths of TCR BV were analyzed by GeneScan technique for detection of the CDR3 size,to evaluate clonality of T cells.Results TCR BV presented specific repertoire skewing in 5 cases in metastatic lymph node,and only 2 ~5 TCR BV subfamilies T cells were identified.Clonal expanded T cells,including monoclonal、oligoclonal and multiclonal patterns.Conclusion There were T cells clonal expansion and selected usage of TCR BV subfamilies in metastatic lymph node of patients with breast cancer,it may be relat-ed to breast cancer associated antigen.
