An histochemical study of regulation of adipogenic and osteoblastic differentiation marrow stromal cells by dexamethasone
Abstract:Objective: To investigate the pathogenetic mechanism of the steroid-induced avascular necrosis of the femoral head,we observed the effect of dexamethasone-regulated marrow stromal cell differentiation into adipocytes. Methods: Marrow stro-mal cells were cultured in the medium with different concentrations of dexamethasone and stained with sudan Ⅲ, adipocyteswere counted. The contents of triglyceride and alkaline phosphatase activity were determined by biochemical assay. Results:The number of adipocyes induced by dexamethasone showed an increase in a dose-time dependent relation. The contents oftriglyceride in experimental groups were as 3.4-times as that of the control, and the alkaline phosphatase activity were de-creased in a dose-dependent relation. Conclusion: Dexamethasone could induced marrow stromal cell differentiation intoadipocytes and inhibite osteoblastic differentiation, which may be related to the pathogenetic mechanism of steroid-osteonecosis.
