Cloning,Bioinformatics and Expression Analysis of TaHsc70-2 Genes in Wheat

Abstract:The heat shock protein gene TaHsc70-2 was cloned from the wheat cultivar Chinese Spring.Subsequently,bioinformatics analysis was performed,and its expression patterns under various hormone treatments[auxin(IAA),gibberellin(GA3),zeatin(ZT),and abscisic acid(ABA)]and stress conditions(drought,salt,high temperature,and low temperature)were analyzed.The results showed that three TaHsc70-2 genes,namely TaHsc70-2-5A,TaHsc70-2-5B,and TaHsc70-2-5D,were cloned,which encoded proteins of 648,647,and 648 amino acids,respectively.The gene structure,protein tertiary structure,and conserved motif arrangements and positions of these three genes were highly similar to those of Hsc70-2 genes in Arabidopsis thaliana and Brachypodium distachyon.The TaHsc70-2 protein sequences were highly conserved,and showed close phylogenetic relationships with orthologous proteins from Hordeum vulgare and Brachypodium distachyon.Protein-protein interaction prediction analysis revealed that TaHsc70-2 proteins had interaction relationships with proteins containing J-domain and HATPase_c domain.The promoter regions of TaHsc70-2 genes contained various cis-acting elements associated with response to hormone,growth and development,and light signaling.TaHsc70-2 genes were expressed in different tissues at various developmental stages of wheat,with relatively higher expression levels in roots.The expression of TaHsc70-2 genes was differential under different hormone treatments,and was upregulated under ZT,high temperature,and drought treatments.It is hypothesized that TaHsc70-2 genes play an important role in wheat growth,development,and responses to abiotic stress.
