Rapid Propagation and Ploidy Identification of Regenerated Plant from Unfertilized Ovary of Cucurbita pepo L.

Abstract:In this experiment,in use of the stem tips of blastocyst regenerated plantlets as explant,the effects of plant growth regulators on the multiplication of shoot tip and the rooting rate of axillary buds were studied,the differences of Cucurbita pepo L.ploidy from root tip,stem tip and tendril tip were compared,and the relationships between chloroplast number,guard cell density and ploidy level of regeneration plants were investigated.The results showed that the optimum medium for axillary buds proliferation and rooting was MS + 0.50 mg/L 6-BA and MS + 0.10 mg/L NAA respectively.The discrepancy of guard cell density and chloroplast number were both significant.The number of stomata per unit area(1.77 mm2)decreased with the increase of ploidy levels,while the number of stomatal chloroplasts increased with the increase of ploidy levels.The plant was assumed to be haploid when the number of chloroplast in stomata guard cell was less than 7,otherwise it was diploid.The accuracies of identification of haploid and diploid plants were 95.73 % and 94.51%.In addition,the tendril tip as a chromosome ploidy identification material is effective,and can be an alternative material of root tip.
