Cloning and Expression Analysis of ZmGLDH Gene in Zea mays L.

Abstract:In order to explore the role of ZmGLDH in maize growth,development and resistance to environmental stresses,primary structure and expression pattern of ZmGLDH gene in response to high salinity were explored by RT-PCR,bioinformatic analysis,and real time fluorescence quantification PCR technology.ZmGLDH gene contains 6 exons and 5 introns.The ORF(open reading frame) of ZmGLDH gene encodes prepropeptide consisting of 39 amino acid (aa)-residue transit peptides,42 aa-residue removal peptide,505 aa-residue mature peptide which included 136 aa-residue FAD-binding domain near N-terminus.ZmGLDH gene was quickly up-regulated and its transcript levels were always higher than that of the control during NaCl treatment.Expression ratio (fold change) of ZmGLDH was increased to 1.20 folds at early stage(1 h),reached a peak(2.31 folds) at 2 h,followed by 2.20 folds at 3 h,and 1.46 folds at 4 h.Results also showed that the up-regulated ZmGLDH expression levels were consistent with higher AsA contents during salt stress,indicating that expression level of ZmGLDH gene is the main factor influencing AsA content in leaves of Zea mays L..
