Antagonistic Activity and Stability of Antifungal Protein of Endophytic Bacterium Y-6 from Zanthoxylum bungeanum against Plant Pathogenic Fungi

Abstract:In order to determine the type and characteristic of antifungal substance synthesized by the endophytic bacterium Y-6 from Zanthoxylum bungeanum and rich the source of bio-pesticide,growth rate method,ammonium sulfate precipitation and SDS-polyacrylamide gel electrophoresis(SDS-PAGE) were used to study the antifungal activity,types,optimal ammonium sulfate saturation and stability.Results showed that secondary metabolites had significant antifungal activity against Alternaria solani,Botrytis cinerea and Alternaria alternate.The inhibitory rates were 90.4%,95.2% and 90.1%,respectively.The antifungal substance was identified as protein and could be completely precipitated at 70% ammonium sulfate saturation.The inhibition zone diameters of the antifungal protein against A.solani and B.cinerea were 24 mm and 26 mm,respectively.Besides,the antifungal protein still had good antifungal activity at pH value of 4-11,below 80 ℃ and after 12 h of UV irradiation.Therefore,it has good potential in the development of bio-pesticide.
