Cloning of Soybean Kunitz T rypsin Inhibitor Gene from Huipizhiheidou and Expression Analysis in Soybean Infected by H eterodera glycines

Abstract：The objective of this study was to research the expression of Kunitz trypsin inhibitor gene in soybean infected by soybean cyst nematode and to clear its function in the resistant mechanism .The soybean Kunitz trypsin inhibitor gene was obtained from roots of Huipizhiheidou (ZDD2315) with high resistance to Heterodera glycines race 3 by RT-PCR ,and sequence analysis indicated that the cloned DNA fragment consisted of 573 bp .The expression of Kunitz trypsin inhibitor gene was detected in the resistant Huipizhiheidou and the susceptible Liaodou 15 infected by Heterodera glycines race 3 by Real-Time Quantitative PCR .The results showed that the relative expression level of Kunitz trypsin inhibitor gene was higher in Huipizhiheidou than in Liaodou 15 at all times ,and it reached the highest in Huipizhiheidou at 20 d after inoculated with Heterodera glycines race 3 ,5 .8 times as high as Liaodou 15 .
