Cloning and Expression Pattern Analysis on ZmCI PK Genes of Maize

Abstract:In order to clone the genes responsing to abiotic stresses in maize ,the primers were de-signed based on the EST sequence of maize CIPK ,and a ZmCIPK gene was cloned from maize (Zea mays) by RT PCR method .The full length of ZmCIPK cDNA was 1 776 bp ,including a 249 bp 5′ UTR ,a 1 350 bp ORF and a 177 bp 3′ UTR .This cDNA sequence encoded a polypepide of 449 amino acid residues with a predicted molecular mass of 50 .64 kDa and a basic isoelectric point of 9 .58 .The deduced amino acid sequence had a high homology with CIPK from Sorghum bicolor .The quantitative real-time PCR result showed that the expression of ZmCIPK gene was induced by abscisic acid ,high salinity ,drought or low temperature .The expression patterns of ZmCI PK under different stresses suggested that this gene might be involved in the response to abiotic stresses .
