Mechanism of Resina Draconis and Indigo Naturalis Compatibility in the Treatment of Diabetic Skin Wound Healing Based on Network Pharmacology

Abstract:Objective To investigate the pharmacological basis and mechanism of Resina Draconis and Indigo natu-ralis compatibility in the treatment of diabetic skin wounds based on network pharmacology.Methods The active ingredi-ents and targets of Resina Draconis and Indigo naturalis were screened by searching the traditional Chinese medicine sys-tematic pharmacology platform(TCMSP)and PubChem.The disease gene database was used to obtain targets related to diabetic skin wounds,the intersection of the active ingredient targets of Resina Draconis and Indigo naturalis with the dis-ease targets of diabetic skin wounds was analyzed as potential targets,and a network of"Resina Draconis and Indigo natu-ralis-active ingredient-potential targets"was constructed.A protein-protein interaction(PPI)network was constructed between the targets,and gene ontology(GO)enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis were performed on the genes of the potential targets.Results Fifteen active ingredients of the Resina Draconis and Indigo naturalis were collected,including diindigodin and indican,and 139 intersection targets were found between Resina Draconis and Indigo naturalis and diabetic skin wounds.Cytoscape network analysis found 10 core targets such as the RAC-alpha serine/threonine-protein kinase(AKT1)and tumor necrosis factor(TNF)in promo-ting diabetic skin wound healing,the targets were mainly enriched in the atherosclerosis signaling pathway,advanced gly-cation end products-receptor for advanced glycation end products(AGE-RAGE)signaling pathway and other related path-ways.Conclusion The Resina Draconis and Indigo naturalis may regulate the healing process of diabetic skin wounds through vascular regeneration,redox,inflammatory response,lipid and atherosclerosis signaling pathways,AGE-RAGE signaling pathway and chemoattractant receptor activation pathways.
