Study on the Construction of High-altitude Hyperuricemia Rat's Kidney Damage Model by Simulating Low-pressure Hypoxic Environment

Abstract:Objective To establish rat model of high-altitude hyperuricemia(HUA)by simulating low-pressure hy-poxic environment and to observe the target organ kidney damage.Methods Male SD rats were randomly divided into blank control group,simple HUA group,high-altitude group and high-altitude HUA group,with 6 rats in each,the modeling process lasted for consecutively 14 d.The rats in the blank control group were placed in the room without inter-vention,rats in the simple HUA group received daily intraperitoneal injections of 250 mg/kg potassium oxonate,rats in the high-altitude group were placed in small animal hypobaric chamber to simulate high-altitude hypoxic environment,rats in the high-altitude HUA group were placed in the hypobaric chamber and received intraperitoneal injections of 250 mg/kg potassium oxonate everyday.After 14 d of modeling,blood samples were collected to test the serum uric acid,creatinine and urea nitrogen levels of rats in each group,their kidney tissues were taken to prepare pathological sections to observe morphological changes.Results After 14 d of modeling,the serum uric acid levels in the simple HUA group,high-alti-tude group and high-altitude HUA group all significantly increased compared to the blank control group(all P<0.01),with the high-altitude HUA group higher than the simple HUA group and high-altitude group(all P<0.01).Compared with the blank control group,the serum creatinine and urea nitrogen levels of rats in the high-altitude HUA group signifi-cantly increased(all P<0.01),and there were obvious pathological changes in the renal tissue structure,while there were no significant abnormalities in renal function indicators in the simple HUA group(P>0.05);com-pared with the simple HUA group,the serum creatinine and urea nitrogen levels of rats in the high-altitude HUA group significantly increased(all P<0.05).Conclusion In this study,we successfully construct high-altitude HUA rat model by simulating low-pressure hypoxic environment combined with intraperitoneal injection of potassium oxonate and find significant target organ kidney damage.This model will help further investigate the potential molecular mechanisms of low-pressure hypoxic environment-induced HUA and its target organ damage and develop special and effective preventive and therapeutic drugs.
