Expression and Clinical Significance of Ubiquitin Specific Proteases 9X and Ubiquitin Specific Proteases 24 in Gastric Adeno-carcinoma Tissue

Abstract:Objective To explore the expression and clinical significance of ubiquitin specific protease 9X(USP9X)and ubiquitin specific protease 24(USP24)in gastric adenocarcinoma(GAC)tissues.Methods The cancer tissues and adjacent normal gastric mucosa tissues of 147 GAC patients who underwent surgical resection in the author's hospital from November 2019 to November 2020 were collected.The positive expression rates of USP9X and USP24 proteins in GAC tissues and adjacent normal gastric mucosa tissues were detected and compared by immunohistochemistry;the rela-tionship between the positive expression rates of USP9X and USP24 proteins and the clinical pathological features of GAC patients was analyzed;the 3-year overall survival rates in USP9X and USP24 positive expression group/negative expres-sion group were analyzed by Kaplan-Meier method;the prognostic factors of GAC patients were analyzed by COX propor-tional hazard model;the relationship between the co-expression of USP9X and USP24 and the clinical pathological fea-tures of GAC patients was analyzed.Results The positive expression rates of USP9X and USP24 proteins in GAC tissues were significantly higher than those in adjacent normal gastric mucosa tissues(P<0.05).Compared to GAC patients with TNM stage Ⅰ+Ⅱ and non-lymph node metastasis,the positive expression rates of USP9X and USP24 in cancer tis-sues of GAC patients with TNM stage Ⅲ and lymph node metastasis significantly increased(P<0.05).Kaplan-Meier analysis showed that the 3-year overall survival rate in GAC patients with USP9X and USP24 positive expressions was significantly lower than those patients with USP9X and USP24 negative expressions(P<0.05).COX regression analysis showed that,lymph node metastasis,positive expressions of USP9X and USP24 and TNM stage Ⅲ were risk factors for the prognosis of GAC patients(P<0.05).Among 147 GAC patients,70 cases(47.62%)were co-expressed with USP9X and USP24,the proportion of USP9X and USP24 co-expressed patients in TNM stage Ⅲ and lymph node metastasis were significantly higher than those of non co-expressed patients(all P<0.05).Conclusion USP9X and USP24 are highly express in GAC tissues,and high expression are related to lymph node metastasis,TNM stage Ⅲand poor prognosis.
