Efficacy and Safety of CCRT Combined with Nimotuzumab for Locally Advanced Nasopharyngeal Carcinoma

Abstract:Objective To investigate the efficacy and safety of concurrent radiochemotherapy(CCRT)combined with Nimotuzumab for locally advanced nasopharyngeal carcinoma(NPC).Methods The randomized controlled trials of CCRT combined with Nimotuzumab in the treatment of locally advanced NPC were searched in Pubmed,Web of Science,EMbase,Cochrane library,VIP database,Wanfang database and China National Knowledge Infrastructure.The Meta analysis of local lesion complete remission,no metastasis survival and adverse event of patients after the combining treat-ment were collected and analyzed by Review Manager 5.4.The local lesion complete remission rate(CRR)and disease control rate(DCR)were set as the primary end point.Results A total of 18 randomized controlled trials,including 1 247 patients.The results of Meta analysis showed that for locally advanced NPC patients,CCRT combined with Nituzumab significantly improved the short-term local lesion CRR of patients compared with CCRT alone(RR=1.34,95%CI:1.24-1.46,Z=7.05,P<0.001)and 1-year metastatic free survival(RR=1.31,95%CI:1.14-1.49,Z=3.96,P<0.001),and significantly improved the DCR of locally advanced NPC patients(RR=1.18,95%CI:1.07-1.30,Z=3.22,P=0.001).Conclusion For locally advanced NPC patients,CCRT combined with Nimotuzumab can improve the short-term local lesion CRR,1-year metastasis-free survival rate and DCR.In addition,it doesn't bring new or aggravate other serious adverse reactions related to CCRT except aggravate skin mucosal reactions in some patients.
