Effects of Poria Cocos Polysaccharide on Intestinal Flora and Liver Function of Mice with Nonalcoholic Fatty Liver Associat-ed Hepatocellular Carcinoma

Abstract:Objective To investigate the effects of Poria cocos polysaccharides(PCP)on intestinal flora and liver function in mice with nonalcoholic fatty liver associated hepatocellular carcinoma.Methods A total of 45 SPF male mice were selected,and 15 were randomly selected as blank group.The blank group mice were fed with ordinary diet,and the other 30 mice were fed with high-fat diet to establish a nonalcoholic fatty liver associated hepatocellular carcinoma mice model,finally,there were 26 mice successfull modeling.The remaining 26 mice were divided into model group(equal amounts of normal saline was given by gavage,n=13)and PCP group(200 mg/kg PCP was given by gavage,n=13)ac-cording to random number table method,with continuously gavage for 4 weeks,and mice in blank group(n=15)were not treated.The pathological tissues,intestinal flora,intestinal flora metabolites,map indexes of denatured gradient gel electrophoresis(DGGE),liver weight,body mass,liver weight ratio,liver function indexes and blood lipid indexes of mice were observed in each group.Results Compared with the blank group,the quantities of Enterococcus,Escherichia coli,Bifidobacterium and lactobacillus and D-lactic acid(D-LAC)of mice in the model group significantly increased,while propionic acid,acetic acid and n-butyric acid significantly decreased,the DGGE map evenness,richness and diversity indexes significantly decreased(all P<0.05);compared with model group,the number of Enterococcus,Escherichia coli,Bifidobacterium and Lactobacillus and D-LAC of mice in PCP group significantly decreased,while propionic acid,acetic acid and n-butyric acid significantly increased,the DGGE map evenness,richness and diversity indexes increased significantly(all P<0.05).Compared with the blank group,liver weight,liver weight ratio,alpha-fetoprotein(AFP),aspartate aminotransferase(AST),alkaline phosphatase(AKP),alanine aminotransferase(ALT),total cholesterol(TC),triglycerides(TG),low density lipoprotein cholesterol(LDL-C)of mice in the model group significantly increased,the body weight and high den-sity lipoprotein cholesterol(HDL-C)significantly decreased(all P<0.05);compared with model group,the liver weight ratio,AFP,AST,AKP,ALT,LDL-C,TC and TG of mice in PCP group significantly decreased,while body weight and HDL-C level significantly increased(all P<0.05).Conclusion PCP can regulate the intestinal flora,improve the lipid metabolism and protect the liver of mice with nonalcoholic fatty liver associated hepatocellular carcinoma.
