Effects of Salidroside on AMPK/SIRT1 Pathway and Lung Function in Rat with Klebsiella Pneumonia Infection

Abstract:Objective To analyze the effect of salidroside(SDS)on adenosine monophosphate-activated protein ki-nase(AMPK)/silenc information regulatory 1(SIRT1)pathway and lung function in rats with Klebsiella pneumonia infec-tion.Methods A total of 60 SPF SD male rats were selected,12 of which were selected as blank group,and the rest es-tablished Klebsiella pneumonia rat model.A total of 44 rats were successfully modeled,and the rats were divided into model group(normal saline was given by gavage,without any other treatment,n=12),SDS low-dose group(5 mg/kg SDS suspension was given by gavage,n=11),SDS medium-dose group(10 mg/kg SDS suspension was given by gavage,n=11),SDS high-dose group(20 mg/kg SDS suspension was given by gavage,n=10).The levels of lung tissue inflammatory factors,lung wet mass index,lung weight,body mass and blood gas index were compared among groups.The protein expressions of AMPK and SIRT1 were detected by Western blot.Results Hematoxylin-eo-sin(HE)staining showed that the lung tissue structure of rats in the blank group was normal,lung pleura was intact,no congestion,no edema interstitium and no inflammatory cell infiltration;compared with the model group,the lung tissue lesions of SDS low-dose group,SDS medium-dose group and SDS high-dose group were significantly reduced.Compared with the blank group,the levels of inflammatory factors,partial pressure of carbon dioxide(PCO2),lung wet mass index in the model group,SDS low-dose group,SDS medium-dose group and SDS high-dose group significantly increased,the levels of arterial oxygen saturation(SaO2),partial pressure of oxygen(PO2),lung weight,body mass,lung volume transformation,airway resistance change,resting ventilation,the expression of SIRT1 and AMPK significantly decreased(all P<0.05);compared with the model group,the levels of inflammatory factors,PCO2 level,lung wet mass index in the SDS low-dose group,SDS medium-dose group and SDS high-dose group significantly decreased,the levels of SaO2,PO2,lung weight,body mass,lung volume transformation,airway resistance change,resting ventilation,the expression of SIRT1 and AMPK significantly increased(all P<0.05);compared with the SDS low-dose group,the levels of inflam-matory factors,PCO2 level,lung wet mass index in the SDS medium-dose group and SDS high-dose group significantly decreased,the levels of SaO2,PO2,lung weight,body mass,lung volume transformation,airway resistance change,resting ventilation,the expression of SIRT1 and AMPK significantly increased,and the reduction was more significant in the high dose group(all P<0.05).Conclusion SDS can reduce the level of inflammation and improved lung function in rats with Klebsiella pneumonia infection,and the mechanism may be related to the regulation of the AMPK/SIRT 1 pathway.
