Mendelian Randomization Study of Primary Biliary Cholangitis and Three Autoimmune Diseases

Abstract:Objective A two-sample Mendelian randomization study was used to investigate the causal relationship between primary biliary cholangitis(PBC)and systemic lupus erythematosus(SLE),rheumatoid arthritis(RA),and sjogren's syndrome(SS)among 3 autoimmune diseases.Methods In the genome-wide association study(GWAS)data-set,the genetic locus closely related to PBC was selected as the instrumental variable,and the causal effect was evaluated by inverse variance weighted(IVW)method.weighted median(WM)and MR-Egger regression were used to supplement the explanation.The analysis results were expressed as OR value and 95%CI.Cochran's Q was used to test heterogenei-ty,and MR-Egger regression was used to test level pleipotency.Results The genetic prediction results suggest a positive causal relationship between PBC and SLE(OR=1.354,95%CI:1.229~1.491,P<0.05),RA(OR=1.097,95%CI:1.061~1.133,P<0.05),and SS(OR=1.388,95%CI:1.223~1.574,P<0.05).MR-Egger regression did not detect potential horizontal pleiotropy(SLE:P=0.62;RA:P=0.56;SS:P=0.76).Conclusion There is a causal association between PBC and SLE,RA,and SS.
