Effect of miR-762 Expression on Proliferation in Ovarian Cancer

Abstract:Objective To evaluate the role of miR-762 on the proliferation of SKOV3 cells.Methods The expressions of miR-762 and menin in 40 patients with ovarian cancer were analyzed by quantitative polymerase chain reaction (qPCR),and the effect of miR-762 on the regulation of menin was further analyzed by luciferase reporter gene assay.After cells were over expressed or silenced of miR-762 cells,the effect of miR-762 on proliferation of SKOV3 cells was observed by methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay.Western blot assay was used to detect the regulatory role of miR-762 in the proliferation of menin and Wnt pathway.Results qPCR detection showed that the expression of miR-762 in tumor was higher than that in adjacent tissues.Western blot and qPCR showed that the expression of menin was lower in ovarian cancer.The report gene experiment proved that miR-762 could directly target menir.MTT detection showed that over expression of miR-762 could promote the proliferation of SKOV3 cells by inhibiting menin and activating Wnt pathway.Conclusion Expression of miR-762 was high in ovarian cancer,and its over expression may promote the development of ovarian cancer by inhibiting menin and activating wnt pathway.
