Analysis of Platelet-related Parameters for Platelet-rich Plasma Preparation by Two-step Centrifugation

Abstract:Objective To analyze the differences of platelet-related parameters in the preparation of platelet-rich plasma (PRP) by two-step centrifugation,discuss the preparation methods of PRP,and provide the theoretical guidance for clinical preparation of qualified PRP.Methods The platelet (PLT) parameters of 56 patients under PRP treatment in department of plastic surgery of authors' hospital were statistically analyzed.The changes of PLT count,mean platelet volume (MPV),platelet distribution width (PDW) and platelet hematocrit (PCT) were detected before centrifugation,after the first centrifugation [1500 rpm/min (377 g),10 min] and the second centrifugation 13000 rpm/min (1509 g),20 min].Results The mean PLT count of PRP,harvested volume and PLT recovery rate after secondary centrifugation were (1016 ± 119) × 109/L,(6.5 ± 1.6) ml,(79.70 ± 9.42) %,respectively.The PLT count,PCT and MPV of whole blood remarkably increased after centrifugation,but PDW reduced significantly.Conclusion PRP can be prepared for clinic by authors' twostep centrifugations.Centrifugation may change in platelet-related parameters,and these changes show that centrifugation may affect the external form,internal structure of PLT,and further influence the quality and function of PRP.
