Mechanism of Proliferation Inhibition of Phloretin in Esophageal Cancer Cell Lines by ERK Signal Pathway

Abstract:Objective To observe the inhibition of phloretin in esophageal cancer cell (EC-109) lines and its mechanism.Methods The inhibition effect of phloretin was detected to EC-109 by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay.The effect of EC-109 apoptosis was tested by flow cytometry.The expressions of Bcl-2-associated X protein gene (Bax) and B-cell leukemia-2 (Bcl-2) protein were detected by enzyme-linked immuno sorbent assay (ELISA).The influence of phloretin in Bax,Bcl-2,extracellular regulated protein kinases (ERK) and phosphorylation of extracelluar signal-regulated kinase (p-ERK) protein expression of EC-109 were detected by Western blot.Results Phloretin obviously inhibited the proliferation of EC-109 in a time and concentration-dependent manner.The flow cytometry results showed phloretin promoted apoptosis of EC-109,and compared with control group,there were significantly different (P<0.05).The results of ELISA and Western blot suggested that phloretin increased the expression of Bax protein in EC-109,inhibited the expression of Bcl-2 protein (P<0.05).Western blot results suggested that phloretin inhibited the expression of p-ERK protein in cells (P<0.05).Conclusion Phloretin inhibit the proliferation of EC-109 and its possible mechanism is through the regulation of ERK signaling pathway.
