Effects of Nicotine on Proliferation and Apoptosis of Mice Bone Marrow-derived Mesenchymal Stem Cells

Abstract:Objective To investigate the effects of nicotine on proliferation and apoptosis of mice bone marrow-derived mesenchymal stem cells (MSCs).Methods Mice MSCs were cultured and identified.Bone marrow-derived MSCs were divided into experimental groups which were treated with different concentration of nicotine and control group without nicotine.The cell viability and apoptosis rate of bone marrow-derived MSCs were detected with terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling (TUNEL) and cell counting kit-8 (CCK-8) under the treatment of different concentration of nicotine (10,50,100 and 200μg/mL) for 24 hours.The expressions of B cell leukemia-2 (Bcl-2),Bcl-2 associated X protein (Bax) and Caspase-3 mRNA were detected by real-time quantitative polymerase chain reaction (PCR).Results The features of the cultured cells fitted MSCs.Compared with control group (0μg/mL),the results of TUNEL and CCK-8 showed cell viability down-regulated in experimental groups (10,50,100 and 200μg/mL) and cell apoptosis rate of MSCs up-regulated (P<0.05).After nicotine treatment for 24 hours,the expression of Bcl-2 mRNA in MSCs was lower than that in control group.The expressions of Bax and Caspase-3 mRNA were higher in experimental groups than those in control group (P<0.05).Conclusion Nicotine can inhibit the proliferation of bone marrow-derived MSCs and promote the apoptosis of MSCs.These results further suggest that smoking may affects the treatment of stem cell therapy on diseases.
