Pathological Effect of Inflammatory Cytokines and Synovium in Rabbits with Osteoarthritis by Meridian-sinew Release Therapy

Abstract:Objective To investgate the effect and mechanism of meridian-sinew release therapy for papain-induced rabbit knee osteoarthritis(OA) on peripheral blood serum,inflammatory factors expression of synovial fluid,and pathological variation of synovial tissue.Methods Twenty New Zealand white rabbits were randomly divided into four groups(n =5 in each group):treatment group(A),control group(B),model group(C),and blank group(D).The rabbits in treatment group,control group and model group were modeled as knee osteoarthritis with the following 4 weeks treatment.The blood and synovial fluid was separately obtained from rabbit ear side vein and knee after the final administration.The contents of interleukin-1β(IL-1 β),tumor necrosis factor(TNF-α),transforming growth factor-β(TGF-β1) and matrix metalloproteinase 13 (MMP-13) in the rabbits' serum and synovial fluid were measured,and the synovial tissues of knee joint cavity were sliced for pathological analysis.Results Compared with model group and control group,levels of IL-1β,TNF-α,TGF-β1,MMP-13 were significantly differences in rabbit peripheral blood and synovial fluid (P<0.05).Control group and treatment group had a significant difference with model group(P<0.05),and there was significant difference between control group and blank group(P<0.05).Conclusion Meridian-sinew release therapy not only reduces effectively the content of IL-1β,TNF-α,TGF-β1 and MMP-13 in rabbits' peripheral blood and synovial fluid but it also lightens the synovial inflammation.
