Abstract:Objective:To analyze the value of aldehyde ketone reductase 1B-10 in predicting hilar cholangiocarcinoma and provide reference for the diagnosis and treatment of this disease. Methods:Selecting 30 patients with hilar cholangiocarcinoma treated in The Second Affiliated Hospital of Nanhua University from April 2024 to October 2024 who were included in the study group,30 healthy individuals who underwent routine physical examinations at the same time in The Second Affiliated Hospital of Nanhua University were included in the healthy group. Both groups were tested for the expression profile of peripheral blood aldehyde ketone reductase 1B-10 using Circular RNAs(circRNAs)microarray chip technology, and the differences were compared to statistically analyze the distribution of alpha-fetoprotein(AFP)in different sample types.Calculating the interference rate of potential interfering tumor markers at different concentrations of tumor markers. Results:Compared with the healthy group,a total of 26 aldehyde ketone reductases 1B-10 in the study group showed fold changes with an absolute value of≥4,of which 14 were upregulated and 12 were downregulated. The distribution of AFP in the research group was mostly 8.1-20.0 and>1 000.0,followed by 0-80.At 20 mAU/mL,AFP,aldo-keto reductase family 1 member B10(AKR1B10),and protein induced by vitamin K absence or antagonist-Ⅱ(PIVKA Ⅱ)had no infection rates. At 200 mAU/mL,AFP interference rate was 4,AKR1B10 interference rate was 1,and PIVKA Ⅱ interference rate was 2.By limiting the analysis of receiver operating characteristic curve(ROC),net reclassification improvement index(NRI),and integrated discrimination improvement(IDI),the area under curve(AUC)of the indicator prediction model significantly increased and the prediction performance improved significantly, with statistically significant differences(P<0.05).Conclusion:The expression of peripheral blood aldehyde ketone reductase 1B-10 is significantly abnormal in patients with hilar cholangiocarcinoma and can serve as a biomarker for clinical diagnosis and treatment.
