Abstract:Objective:To investigate the effects of low, medium, and high doses(100 mg/kg,200 mg/kg,300 mg/kg)of saponins from Gynostemma pentaphyllum on oxidative stress levels in a rat model of chronic nonbacterial prostatitis(CNP).Methods:A total of 70 specific pathogen free(SPF)grade SD rats were selected, with 15 rats as the blank group and 55 rats as the model group.Establishing a CNP rat model using carrageenan after one week of modeling,5 rats from each group were euthanized to confirm the modeling results. After successful modeling, the model rats were divided into five groups:model group,Qianlietai capsule group, and low, medium, and high dose groups of Gynostemma pentaphyllum saponins, with 10 rats in each group. After 4 weeks of drug intervention, the differences in prostate index(PI),superoxide dismutase(SOD),malondialdehyde(MDA),and glutathione peroxidase(GSH-Px)content were observed in each group of rats. Results:The PI of the Qianlietai capsule group and the low, medium, and high dose groups of Gynostemma pentaphyllum saponins were significantly reduced compared to the model group, the difference was statistically significant(F=46.586,P<0.01).Compared with the model group, the levels of SOD and GSH-Px in Qianlietai capsule group and the low, medium, and high dose groups of Gynostemma pentaphyllum saponins were significantly increased, while the levels of MDA were significantly decreased, and the difference was statistically significant(F=219.959,21.406;P<0.01).Compared with the Qianlietai group, the high-dose group of Gynostemma pentaphyllum saponins showed significantly increased levels of SOD and GSH-Px,and significantly decreased levels of MDA,with statistically significant differences(F=96.252,1.459,2.899;P<0.01).Conclusion:Low,medium,and high doses of Gynostemma pentaphyllum saponins could significantly reduce prostate index, while medium and high doses could increase SOD and GSH-Px levels, reduce MDA levels, thereby exerting antioxidant effects and delaying disease progression
