Rare cisAB Blood Type Phenotype and Genotype Identification and Family History Investigation

Abstract:Objective:To identify 1 case of a difficult ABO blood type in an unpaid blood donor and conduct a family investigation.Methods:Firstly,the ABO blood group was identified by serological method,and then the gene analysis was carried out by molecular biological method.Results:The proband and his father both had c.467C/T and c.803G/C point mutations in the 7th exon of the ABO gene,with a genotype of cisAB.01/O.01 and a phenotype of the cisAB subtype.His mother and brothers were all normal O type.The proband's cisAB.01 allele was inherited from his father.Conclusion:The cisAB.01 allele causes red blood cells to express normal A antigen and weaker B antigen.Serological methods for identifying ABO subtypes present certain difficulties,but molecular biology methods can accurately identify specific subtypes.
