Analysis of HPA Gene Typing in Platelet Transfusion Recipients and Gene Database Capacity

Abstract：Objective:To investigate human platelet-specific antigen(HPA)genotyping and regional polymorphism among platelet transfusion recipients in Zhongshan,while analyzing the capacity level of the HPA gene database,thereby providing data support for HPA gene matching among platelet transfusion patients in this region.Methods:Genotyping of the HPA-1 to 6/10/15/21 alleles was performed in 135 platelet transfusion recipients in Zhongshan.HPA gene frequencies and haplotype frequencies were calculated.Based on the phenotypic frequencies,the probability of finding a fully matched HPA donor for platelet transfusion recipients in Zhongshan was calculated to assess the optimal blood bank capacity level.Results:Polymorphism data for the HPA-1 to 6/10/15/21 phenotype groups among platelet transfusion recipients in Zhongshan region were obtained.The allele frequencies for each system were as follows:1a∶0.996 3,1b∶0.003 7,2a∶0.974 1,2b∶0.025 9,3a∶0.544 4,3b∶0.455 6,5a∶0.974 1,5b∶0.025 9,6a∶0.988 9,6b∶0.011 1,15a∶0.529 6,15b∶0.470 4.HPA-4,10,21 system results showed monospecificity.Calculations indicated that when the platelet donor pool capacity was 403 individuals,patients had a 95%probability of finding at least one donor with full HPA-1 to 6/10/15/21 matching within that pool.Conclusion:The distribution of HPA genotypes among platelet transfusion recipients in Zhongshan exhibits unique local polymorphisms.When the platelet donor pool reaches a capacity of 403 individuals,it can largely meet the demand for finding HPA-matched donors for patients in this region who have experienced ineffective platelet transfusions.
