Effects of Total Saponins of Panax Ginseng on Blood Lipids and Oxidative Stress Levels in AS Rats

Abstract:Objective:To study the effects of Panax ginseng total saponin(PNS)on blood lipids,serum superoxide dismutase(SOD),malondialdehyde(MDA),superoxide anion(O2-)and hydroxyl radicals(·OH)in atherosclerotic(AS)rats.Methods:A total of 60 healthy male SD rats were used,which were divided into blank control group(Group A),atherosclerosis model group(Group B),PNS low dose group(Group L),PNS medium dose group(Group M),PNS high dose group(Group H),and atorvastatin group(Group Y),with 10 rats in each group,by the random allocation method.The high-fat diet+intraperitoneal injection of VD3 method was used to intervene for 8 weeks to carry out the establishment of AS rat model.At the end of the intervention,1 rat was randomly selected from each group to confirm the success of atherosclerosis modeling.After successful modeling,the drug was administered by gavage to groups B,L,M,H,and Y for 8 weeks.After 4 weeks of drug intervention,the liver index,atherosclerotic index(AI),serum lipid levels,endothelial diastolic factor(EDF),nitric oxide(NO),SOD,MDA,O2-,and·OH were measured in the rats.Results:Compared with group A,the levels of total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),and hepatic index of serum in group B were significantly increased,and the difference was statistically significant(P<0.01).The ability to inhibit O2-and·OH,and total SOD activity decreased,and the difference was statistically significant(P<0.05).Compared with group B,group L was able to improve the level of LDL-C in serum with statistically significant difference(P<0.01),while the antioxidant effect on As was not significant.While serum TC,TG,LDL-C,AI1,AI2,and MDA levels were significantly reduced in groups M and H,the content of high-density lipoprotein cholesterol(HDL-C),the vitality of T-SOD,and the ability to inhibit O2-were sequentially elevated,and the content of NO was improved,and the difference was statistically significant(P<0.05).Conclusion:The PNS retard the formation of AS through hypolipidemic and antioxidant effects.
