Regulation of ZNF148 on Insulin Secretion of Rat Pancreatic β Tumor Cell Lines

Abstract:Objective:To investigate the regulatory effects of Zinc finger protein 148(ZNF148)on insulin secretion of rat pancreatic β tumor cells.Methods:RNA interference technique(siRNA)was used to knockdown the ZNF148 gene.The protein expression level of ZNF148,ANXA 2 and VNUT were examined by Western blot.The activity of pancreatic cells after ZNF148 interference was detected by CCK-8 and the concentration of insulin in cell supernatant was detected by ELISA.Results:After knockdown of ZNF148,the expression of ZNF148 was significantly decreased,the ANXA2 and VNUT were significantly increased the difference were statistically significant(P<0.05);CCK-8 results showed that no difference in significant statistically effect on cell activity of normal cell lines(P>0.05);the insulin secretion function was significantly increased(P<0.05).Conclusion:ZNF148 is a Krüpple-like Zinc finger transcription factor in INS-1 cell,ZNF148 knockdown has no effect on cell activity,but significantly enhanced the expression of ANXA 2 and VNUT gene,suggesting that ZNF148 may play an important role in the development of type 2 diabetes,which might be a potential new target for type 2 diabetes targeting therapy.
