Expression of ALKBH5 in Thyroid Cancer and Its Clinical Significance

Abstract：Objective:To clarify the expression of N6-methyladenosine(m6A)demethylase ALKBH5 in thyroid carcinoma tissue and blood and its relationship with clinicopathological features and prognosis,so as to provide insights for the prevention and treatment of thyroid carcinoma.Methods:Quantitative real-time polymerase chain reaction(qRT-PCR)was used to detect the expression of ALKBH5 mRNA in the peripheral blood.Immunohistochemical experiments were used to detect the expression of ALKBH5 protein in 58 pairs of thyroid carcinoma tissues and thyroid paracancerous tissues.Using bioinformatics method,Kaplan Meier plotter online database was used to analyze the effect of ALKBH5 expression on the prognosis of patients with new-onset thyroid cancer.Results:The results of qRT-PCR experiments showed that the expression level of ALKBH5 mRNA in the blood of patients with thyroid carcinoma was significantly up-regulated,the difference was statistically significant(t=2.719,P＜0.05).The results of immunohistochemistry showed that the high expression of ALKBH5 accounted for 96.55%in thyroid carcinoma tissues,higher than that 12.07%in adjacent tissues,and the expression difference was statistically significant(χ2=52.841,P＜0.05).Logistic regression analysis showed that in patients with thyroid carcinoma,the high expression of ALKBH5 was related to age,TNM stage,and clinical stage(χ2=14.550,4.852,4.580,P＜0.05).The survival rate of new thyroid carcinoma patients with high expression of ALKBH5 was poor,and the difference was statistically significant(P＜0.05).Conclusion:Compared with adjacent tissues,the expression of ALKBH5 in thyroid carcinoma tissues and blood was significantly increased,and it was related to patient age,TNM stage,clinical stage and survival time.The survival rate of new thyroid carcinoma patients with high expression of ALKBH5 is poor.
