Study on TCF3 Controlling Proliferation and Invasion of Endometrial Cancer Cells

Abstract:Objective:To investigate the expression pattern of transcription factor 3(TCF3)in endometrial cancer and its impact on the proliferative and invasive capabilities of cancer cells.Methods:The expression of TCF3 in endometrial cancer was analyzed using the TCGA database,along with its correlation to patient prognosis.RNA interference technology was employed to knock down TCF3,and the effects on endometrial cancer cell biology were assessed using CCK8 proliferation assays,Transwell invasion assays,and flow cytometry for apoptosis detection.Results:The expression level of TCF3 in EC tissues was significantly higher than that in adjacent tissues(Kruskal-Wallis chi-squared=30.53,P<0.05).High expression of TCF3 was closely related to the poor prognosis of patients(logrank test,Chi-square statistic=11.63,P<0.05).The t test showed that knockdown of TCF3 significantly inhibited cell proliferation(P<0.001)and invasion ability(P<0.001),and promoted cell apoptosis(P<0.05).Cell cycle analysis showed that knockdown of TCF3 did not cause significant changes in the cell cycle.Conclusion:TCF3 promotes cell proliferation and invasion in endometrial cancer,and these effects are independent of cell cycle regulation,suggesting that TCF3 may serve as a potential therapeutic target for endometrial cancer.
