Hamster Buccal Tumors Induced by DMBA Implantation Under Hamster Cheek Pouchs and Establishment of Their Derived Cell Line HamOC

Abstract:Objective:To induce a hamster buccal tumor model using the carcinogen DMBA,and establish a hamster buccal tumor derived tumor cell line.Methods:7,12-dimethylbenzanthracene(DMBA)solution was absorbed by A circular filter paper with a diameter.Under anesthesia and sterile conditions,the hamster cheek pouch was open,for entering the filter paper with DMBA under the cheek pouch mucosa,then the wound was sutured,and the filter paper was fixed to a specific part of the cheek pouch with silk thread.60 days after operation,the hamsters were sacrificed to examine the tumor formation.Tumor tissues were taken for the in vitro culture of primary cell lines using primary cell culture techniques.The growth,proliferation and nude mouse implantation of the cell lines were observed.Results:Using the method of embedding DMBA under the mucosa of hamster cheek pouches,an animal model of hamster buccal tumor was induced.Through pathological analysis of the hamster buccal tumor,HE staining showed that the tumor was a undifferentiated carcinoma.The immunohistochemical results showed Vimentin positive staining and E-cadherin negative staining.A hamster oral cancer-derived cell line(hamOC)was obtained through primary culture of hamster buccal tumors.When hamOC cells were inoculated subcutaneously into the back of hamsters and subcutaneously into nude mice,and transplanted tumors could be formed in both cases,suggesting that this cell line is tumorigenic.The pathological section of the transplanted tumor showed undifferentiated carcinoma,positive for Vimentin and negative for E-cadherin.Conclusion:A hamster buccal tumor model is successfully established using the improved DMBA induction method,and a hamster oral cancer cell line hamOC is cultured,which conforms to the characteristics of malignant tumor cell line and can be used as an experimental cell model for oral cancer research.
