Comparison of TAC Technology and qPCR Method in Rapid Detection of Various Respiratory Pathogens

Abstract:Objective:To compare the application value of TaqMan array cards(TAC)technology and quantitative real-time PCR(qPCR)method in rapid detection of various respiratory pathogens.Methods:A total of 126 clinical samples of patients with respiratory tract infection who were treated in the outpatient department of respiratory medicine of the hospital from June 2022 to June 2023 were collected.TAC technology and qPCR method were used to detect respiratory pathogens,and the detection value of the above two methods for multiple respiratory pathogens was analyzed.Results:TAC technology showed that 102 of 126 samples were positive,with a positive rate of 80.95%(102/126),including 16 cases of rhinovirus,12 cases of respiratory syncytial virus,16 cases of adenovirus,18 cases of parainfluenza virus,14 cases of mycoplasma pneumoniae,12 cases of influenza A virus and 14 cases of influenza B virus.Among 126 samples,94 were positive by qPCR,with a positive rate of 74.60%(94/126).Among them,16 were rhinovirus,10 were respiratory syncytial virus,13 were adenovirus,16 were parainfluenza virus,16 were Mycoplasma pneumoniae,10 were influenza A virus,and 13 were influenza B virus.Among 126 samples,114 cases showed consistent results between the two methods,with a positive consistency rate of 73.02%(92/126)and a negative consistency rate of 17.46%(22/126).Only 12 samples showed inconsistent test results.Among 12 samples with inconsistent detection results between the two methods,8 samples were consistent with TAC detection results after review,including 6 positive results and 2 negative results.After review,the results were consistent with the results of qPCR in 4 samples,including 2 positive results and 2 negative results.The positive coincidence rate and total coincidence rate of TAC technology and qPCR method in the detection of rhinovirus,respiratory syncytial virus,adenovirus,parainfluenza virus,Mycoplasma pneumoniae,influenza A virus and influenza B virus were all>80%,and the Kappa value was between 0.634 and 1.000,indicating that the results of TAC technology and qPCR method had high consistency.Conclusion:TAC technology and qPCR method have good application value in the rapid detection of various respiratory pathogens,and these 2 methods have high consistency in detecting respiratory pathogen types.
