Relationship of Gene Polymorphism of MTHFR rs1801131 with Susceptibility and Severity of Chronic Obstructive Pulmonary Disease

Abstract:Objective:To investigate the relationship of the gene polymorphism of MTHFR rs1801131 with susceptibility and severity of chronic obstructive pulmonary disease(COPD).Methods:The geneotype and allele distributions of MTHFR rs1801131 gene were detected in 200 COPD patients group and 200 healthy control group by the polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).The automatic biochemistry analyzer and lung function meter were used to detect the biochemistry and lung function.Logistic regression was used to analyze the influencing factors of COPD,and the genotype distribution of COPD patients with different severity was analyzed.Results:The levels of FEV1/FVC,FEV1%pred and FOL in COPD group were lower than those in control group,while the levels of smoking,TG,TC,LDL,HDL and Hcy in COPD group were higher than those in control group.The differences were statistically significant(t=1.333,-37.364,-38.810,-12.640,6.360,11.868,8.439,3.083,11.082;P<0.05).There were significant differences in genotype frequencies of AA,AC and CC and allele frequencies of A and C of MTHFR rs1801131 between the two groups(χ2=6.511,5.780;P<0.05).The Hcy level of MTHFR rs1801131 was compared among different genotypes in COPD group,and the difference was statistically significant(P<0.05).There was no significant difference in MTHFR rs1801131 gene frequency in patients with different severity of COPD(χ2=0.483,0.074;P>0.05).COPD severity grade was positively correlated with Hcy level,COPD severity grade was negatively correlated with FOL level,and Hcy level was negatively correlated with FOL level(r=-0.253,P<0.001).The results of logistics regression analysis showed that smoking,Hcy,FOL and MTHFR rs1801131 gene polymorphisms were the influencing factors of COPD.Conclusion:Smoking,Hcy,FOL and MTHFR rs1801131 gene polymorphisms are risk factors for COPD.MTHFRrs1801131 has no significant relationship with the severity of COPD,and MTHFRrs1801131 may be a genetic susceptibility gene for COPD.
