Effect of High-dose rHGH Combined with Lysine Dicalcium Phosphate Granules on Bone Metabolism and Growth and Development in Children with Short Stature

Abstract:Objective:To observe the effects of high-dose recombinant human growth hormone(rHGH)combined with lysine dicalcium phosphate granules on bone metabolism and growth and development in children with short stature.Methods:100 children with idiopathic short stature(ISS)who were diagnosed and treated in the hospital from January 2020 to December 2021 were selected as the study subjects.They were randomly divided into an observation group and control group through random number table method,with 50 cases in each group.The control group was treated with lysine dicalcium phosphate granules+conventional dose[0.15 U/(kg·d)]rHGH,while the observation group was treated with lysine dicalcium phosphate granules+high-dose[0.20 U/(kg·d)]rHGH for 12 months.The growth hormone indicators of insulin like growth factor 1(IGF-1)and insulin like growth factor binding protein 3(IGFBP-3),bone metabolism indicators[osteocalcin(OC),bone alkaline phosphatase(BAP),type Ⅰ collagen cross-linked carboxy terminal peptide(Ⅰ-CTP)]and growth and development indicators[height,height standard deviation score(Ht SDS),weight,and growth rate]were compared between the two groups of children before treatment and at 12 months of treatment.The incidence of adverse reactions in both groups of children during the treatment period was calculated.Results:After 12 months of treatment,the levels of serum IGF-1 and IGFBP-3 were significantly increased in both groups,and the levels of serum IGF-1 and IGFBP-3 were higher in the observation group(t=9.460,4.284;P<0.05).The levels of serum OC,BAP,and Ⅰ CTP were significantly increased in both groups,and the above indicators were higher in the observation group(t=6.798,2.794,9.981;P<0.05).The height,Ht SDS,weight,and growth rate of both groups increased,and the above indicators were higher in the observation group(t=4.049,6.044,2.236,13.271;P<0.05).During the treatment period,there was no statistically significant difference in the incidence of adverse reactions between the two groups(x2=0.379,P>0.05).Conclusion:High dose rHGH combined with lysine phosphate calcium granules can improve the levels of growth hormone and bone metabolism in children with ISS,promote their growth and development,and have good safety.
