Effect of Oxymatrine on Action Potential Duration of Cardiac Moy cytes in Guinea Pigs

Abstract:Ab srt act:Objective To observe the effect of oxymatrine on action potential of cardiac myocytes in guinea pigs , and reveal the ionic antiarrhythmic mechanism of oxymatrine .Methods Single cardiac myocytes were isolated by enzyme digestion method , and whole -cell path-clamp technique was used to study the effect of oxymatrine on action potential. Re sults In cardiac myocytes of guinea pig , oxymatrine decreased action potential duration ( APD), without effect on the action potential amplitude and resting potential ( RP).After oxymatrine, APD50(1 μmol· L-1) decreased from (534.7 ±29.62)ms to (490.3 ±47.39)ms (n=5, P<0.01), and APD90 from (572.1 ±49.98)ms to (549.8 ±35.42)ms (n=5, P<0.01).After washout, APD50 returned to (498.1 ±31.78)ms (n=5, P<0. 01) and APD90 returned to (554.2 ±61.28)ms (n=5, P<0.01).Conclusion Oxymatrine could decrease action potential duration .It is suggested that the possibly mechanism may be related to the ionic antiarrhythmic effect of oxymatrine .
